Without a programming background, implementing such techniques can be daunting, and manual coding via direct observation often remains a preferred alternative among researchers, particularly for small-scale projects (Gomez-Marin, Paton, Kampff, Costa, & Mainen, 2014). Methods for automated tracking abound in the computer vision literature, yet researchers often lack the know-how to implement these solutions for their specific tracking needs. Attanasi et al., 2014 Mersch, Crespi, & Keller, 2013). Such automated tracking techniques drastically reduce the time and effort required to extract image-based data, allowing researchers to generate large and highly detailed datasets of animal movement and behaviour (e.g. The field of computer vision now allows researchers to automatically track subjects in a variety of contexts (Dell et al., 2014). As such, researchers in diverse fields of science require methods allowing noninvasive measurements of movement, in different environments, across many animals simultaneously, and with high precision and accuracy (Dell et al., 2014). Movement is also increasingly used as a proxy to evaluate sub-lethal effects of pharmaceuticals, climate change and other anthropogenic stressors on animals, thereby improving our understanding of contemporary environmental and health issues of global importance (Brodin, Fick, Jonsson, & Klaminder, 2013 Hellström et al., 2016 Sundin, Jutfelt, Thorlacius, Fick, & Brodin, 2019). Quantifying animal movement is central to answering numerous research questions in ethology, behavioural ecology, ecophysiology, biomechanics, neuroscience and evolutionary ecology. Tracktor is code-based but requires no coding skills other than the user being able to specify tracking parameters in a designated location, much like in a graphical user interface.

Fiji users only need to enable the ImageScience update site to install MTrackJ. The plugin can handle up to five-dimensional (5D) images of any type supported by ImageJ. MTrackJ is an ImageJ plugin to facilitate tracking of moving objects in image sequences and the measurement of track statistics. MTrackJ is an ImageJ plugin to facilitate manual tracking of moving objects in image sequences and the measurement of basic statistics of traveled distances and velocities.

MANUAL TRACKING IMAGEJ UPGRADE
To install ImageJ 1.49 on a computer with Java pre-installed, or to upgrade to the Tiago Ferreira's comprehensive ImageJ User Guide is available as an 8MB. The ImageJ ‘Chemotaxis and Migration Tool’ plug-in was used to quantitate data (see the section on analysis). The data were exported as multipage TIFF files to enable manual cell tracking using the ImageJ ‘Manual Tracking’ plug-in. Photographs were taken at 5 minute intervals.
